Abstract

The success of nanoparticle drug delivery depends on the ability of the therapeutics to reach their designated cellular and intracellular target sites, while minimizing accumulation and action at nonspecific sites. They cannot freely diffuse through the blood-brain barrier (BBB) and require receptor-mediated transport through brain capillary endothelium to deliver their content into the brain parenchyma. This review presents their general properties of BBB and pharmacokinetics of drug delivery and currently available methods for nanoparticle preparation. Strategies for special formulation that enable drug transport through the blood brain barrier arise from knowledge of the molecular and cellular biology of BBB transport processes.
